While mothers appear in many works, if you expand the scope to mother-like figures, there were likely quite a few candidates.
However! Shining in 1st place with an overwhelming 46.2% of the vote is MOTHER!
This masterpiece, created by copywriter Shigesato Itoi, is well-known for its catchphrase, "No crying until the end," and its iconic solid red package design.
While it helps that the title is literally MOTHER, you will understand its true meaning once you actually play the game and reach the ending!

Coming in 2nd place with 16% of the vote is Dragon Quest III: The Seeds of Salvation, which is also the title I voted for!
In addition to being a legendary title, many people likely have the opening scene strongly burned into their memories.
The most unique entry among them is likely Touhouken Bunroku in 3rd place.
